DescriptionL189 is a DNA ligase I, III and IV inhibitor (IC50 values are 5, 9 and 5 μM respectively) that blocks DNA binding. L189 inhibits base excision repair (BER) and non-homologous end joining (NHEJ). L189 specifically sensitizes cancer cells to DNA damage and increases the cytotoxicity of DNA-damaging agents. L189 will not only provide insights into the cellular function of these enzymes but also serve as lead compounds for the development of anticancer agents.

In VitroL189 inhibits DNA Ligase I, III and IV activity with IC50 values of 5 μM, 9 μM and 5 μM, respectively.L189 (5 μM, 48 h) bring good anti-proliferation activity preferentially and cause cell death by creating a cytotoxic environment.L189 (5 μM, 48 h) reduces HeLa nuclear staining along with TMZ.L189 (5 μM, 48 h) enhances TMZ-induced HeLa growth arrestwhen along with TMZ that possibly in G2/M cell cycle phase without employing cell death mechanisms.
